Number One

img15372hf.jpg

To avid Bleach fans out there, you know what song this is. You know when Ichigo made his appearance this song will be accompanying him. You know that's when all the machoism and coolness ooze into our senses (no kidding)! here's the lyrics for Number One by Hazel Fernandez. Shine on!

If you wanna see some action
Gotta be the centre of attraction
Make sure that they got their eyes on you, like the face that you see on every magazine

Be the focus of attention
Be the name that every one must mention
Come out from the shadows, it's your time, cos tonight is the night for everyone to see

It’s natural
You know that this is where you gotta be, it must be your destiny
Sensational
And you believe that, this is what you've waited for and it's you that they all adore, so baby

Now you feel like number one
Shining bright for everyone
Living out your fantasy, the
Brightest star for all to see
Now you feel like number one
Shining bright for everyone
Living out your fantasy, you're the
Brightest star there's ever been

Feel the heat that's all around you
Flashing lights and ecstasy surround you
Everybody wants a piece of you; you're the queen of the scene, living in a dream

It’s natural
You know that this is where you gotta be, it must be your destiny
Sensational
And you believe that, this is what you've waited for and it's you that they all adore

Now you feel like number one
Shining bright for everyone
Living out your fantasy, the
Brightest star for all to see

This is what you've waited for and it's you that they all adore

Now you feel like number one
Shining bright for everyone
Living out your fantasy, the
Brightest star for all to see
Now you feel like number one
Shining bright for everyone
Living out your fantasy, you're the
Brightest star there's ever been

So4mo

1 more hour to conclude my first day as a sophomore.

First thing in the morning i watered a small pot of kalanchoe next to my bed. I bought it out of the blue coz my green fingers are itching. Then after bathing, i took a brisk 15 minutes' walk to Lilly hall. The whole campus is alive with so many people! The whole vibrant is highly contagious. I walked into my research lab and dropped some bak zhang for my professor and her lab members as promised for taking care of me during my internship.

First lecture started at 9.30am. The whole lecture hall is filled to the brim! Old friends met for the first time after summer, new friends made sitting at the next seat, new professor cracking up new jokes and giving new instructions for the new semester... I met up with David, a korean guy, and spent a few minutes catching up with each other. The fact that i have been to that lecture hall for the 3rd course now makes me feel old T.T Professor Hollenback, who happens to be my research professor's husband, is an amiable guy. He happens to be a left-hander too, and had problems with the made-for-right-hander projector. His hands were always over the words that he wrote on the tranparency, but unlike the others I found that i wasn't annoyed at all.

The next class is japanese class. After everyone has taken their seat, Fukuda-sensei spoke in rapid japanese straightaway. We were all dumbfound. Then she switched on powerpoint and wrote in English, 'no english allowed during class, only polite requests are acceptable'. So we had to pick up the language like a baby. I think i need to use the polite request 'eego de hanashitemo iidesuka?' (may i speak to you in english?) A LOT. But overall we had fun. Fukuda-sensei was teaching us how to address peers by adding -san behind their last name, and to give us a more realistic example she used celebrity photos. So we knew (ala japanese slang) Nicole Kidman is Kidoman-san, Tom Hanks is Hankes-san, Katie Holmes is NOT Cruise-san but still Holmes-san because Tom finds marrying Katie a 'mission impossible' etc etc. Eventhough Fukuda-sensei was unable to articulate that juicy piece of gossip in english, we all understand well without any translations :P

2 more classes of bio lab and organic chemistry and i was done for the day. Time to shop for books. Came out of bookstore with my wallet a lot thinner. i spent about $500 up till now for 3 textbooks, 3 lab manuals and 1 workbook. It's really expensive, but i was lazy to find a better deal on the net, and plus it takes a longer time to get it delivered.

Shortly returning to my dorm room while i was setting up the internet connection a junior came by to chat. He's from Penang Chung Lin just fresh out from SPM. He was having a bad first day, so being a good senior (:P) i lent him my ear and advised him.

Had a very big dinner at the dining hall afterwards. Man i feel like this whole year is fit to be called a 'delayed-freshman 15' syndrome. (to the uninitiated, 'freshman 15' is a common 'disease' among freshmen who spurge in buffet-style dining halls and ultimately gain 15 pounds in the first year. I wasn't dining hall-fed in freshman year)

More japanese class tomorrow! And bio experiment starts tomorrow too! Very looking forward to attending them..

Presentation

I'm writing a script for my presentation. Please ignore me.

39-1.2B.r2s4.10x.Slit2-3A10.gif

Archive no: 5
File Name: 39-1.2B.r2s4.10x.Slit2-3A10
Embryonic day: 4

Script:

This is a microscope imaging of part of the chick inner ear of an E4 embryo. At this age the ear is still a globular structure called the otocyst. With cell lineage analysis we can precisely determine the development pathway for different areas of presensory cells, but for my exercise i'm learning how to identify the anatomy of the chick inner ear.

*eye contact, eye contact*

Now we are looking at the ventral view of the left otocyst (point on the projector screen) located at the medial-posterior side of the image. This section is double-stained with Slit2 and 3A10, the former expressed in blue color and the latter, brown.

We are actually now looking at the more dorsal section of the otocyst, so what we have here, I deduce from previous papers, are the presensory areas of the anterior cristae (point at ac) and posterior cristae (ditto at pc). To further prove this, the adjacent section to this one (switch to next image) which is stained with Bmp4 shows both areas that are presumed to be ac and pc are expressing Bmp4. In Laura Cole et all paper, "Sensory Organ Generation in the Chick Inner Ear: Contributions of Bmp4, Ser1 and L-fng", there are 2 Bmp4 images of Stage 23/24 embryo... oh my embryo is staged at 21.. which shows morphologically similar areas in the otocyst which are stained. Therefore i dare to suggest that the ac and pc are correctly labeled.

The endolymphatic duct (point at ed) is a non sensory part of the the otocyst which will function as a connector to transfer fluid from endolymphatic sac to the hollow regions in a fully developed ear. But is it possible that the ed is expressing Slit2? I'm not very sure about that. Or maybe that is not the endolymphatic duct. I only deduce it from the Cole paper. The only analysis that i can explain is that... alamak what can i say now... hmm.. okay talk about the neuronal midline.. The neuronal midline has a beautiful expression of Slit2.. This is most famously known from the study of Slits and RObos in Drosophila Melanogaster. In Drosophila, the midline expresses strong signal of Slit2 to repel Commissureless axons after crossing the midline to prevent the axons form recrossing. The image is insufficient to show the same conclusion in the case of chick inner ear, but from the expression of 3A10, which shows neurofilaments stained in brown color, there are abundant of axons crossing the midline, regulated by Slits and Robos.

Hmm.. enough for today la. Tiredness...
